Used in various biological application such as electrophoresis protein separation, Tris effective buffer range between pH7.0 and 9.1. In general to work with other buffers to provide a stable pH range. | ||||
Description:
Tris (CAS#77-86-1), also known as tris(hydroxymethyl)aminomethane or tris(hydroxymethyl)aminomethane, is commonly referred to as the second-generation Good's buffer. It is a white crystalline substance that is easily soluble in water and has no irritating odor. Compared to traditional Good's buffers, Tris has better solubility and stability.
Application:
Tris is a very important biological buffer, with Tris HCl is a commonly used buffer salt pair that provides a stable weak alkaline environment (pH 7.0-9.1) and is sensitive to temperature and concentration.
The pH of Tris alkaline solution is around 10.5, and hydrochloric acid is generally added to adjust the pH to the desired value to obtain a buffer solution with that pH value.
People often add EDTA to Tris hydrochloric acid buffer solution to make "TE buffer", which is used for the stability and storage of DNA. If the acid solution used to adjust the pH value is replaced with acetic acid, "TAE buffer" (Tris/Acetate/EDTA) is obtained, and if it is replaced with boric acid, "TBE buffer" (Tris/Borate/EDTA) is obtained to provide a stable pH buffering environment for reagents.
