PC-300 Preservative

PC-300 preservative is a widely used bacteriostatic agent for in vitro diagnostic reagents. Its main active ingredients are a mixture of two isothiazolinones: 5-Chloro-2-methyl-4-isothiazolin-3-one (CMIT) and 2-Methyl-4-isothiazolin-3-one (MIT), with a content ratio of 3:1, and the total concentration of this mixture is 3.0–3.6%.

It is widely applied in biochemical reagents, immunoassays, coagulation reagents, molecular biology reagents, electrophoresis buffers, PCR/DNA detection-related buffers, hematology reagents, various diluents, buffers, and washing solutions, among others.


Antimicrobial Mechanism of PC-300

The two main active components share the same mechanism: inhibiting cell growth and inducing cell apoptosis. Within minutes of contact with the cell membrane, the active ingredients penetrate into the cell and inhibit intracellular enzyme activity.
The target enzymes are central to the Krebs cycle (cellular metabolism). PC-300 acts on four distinct sites in the Krebs cycle:
  • Pyruvate dehydrogenase

  • α-Ketoglutarate dehydrogenase

  • Succinate dehydrogenase

  • NADH dehydrogenase

This inhibits cell growth, metabolism, and macromolecular synthesis, causing a rapid drop in intracellular energy levels. As the energy system collapses, cells can no longer synthesize compounds required for routine metabolism. Since all bacteria and fungi possess at least part of the Krebs cycle, Niusha PC-300 has a very broad spectrum of activity.

Product Performance & Notes

NeoCide PC-300 preservative is equivalent to Proclin 300. It is essentially consistent with Proclin 300 in HPLC testing, antimicrobial efficacy, and other performance indicators, making it an excellent alternative to Proclin 300.
Due to its inherent product characteristics, the following precautions should be observed during application:
  1. NeoCide PC-300 features a dual bacteriostatic system, offering stronger antimicrobial activity, rapid bactericidal effect, long-term stability, and a wide application range, making it a preferred choice.

  2. It is not suitable for reagents with pH > 8.5. For such formulations, we recommend NeoCide PC-950, PC-950Plus, GML-5, BIT-10, or other compatible preservatives.

  3. High concentrations of PC-300 are not recommended in concentrated buffers, as they may cause yellowing or precipitation. In concentrated formulations, use a low level of PC-300 (e.g., ≤0.05%) combined with primary preservatives such as NeoCide PC-950, PC-950Plus, BND-10, BIT-10, GML-2, GML-3, and GML-5, which do not cause yellowing.

  4. In some applications, PC-300 used alone may show insufficient antimicrobial effect. Simply increasing the concentration of PC-300 may not improve efficacy. Instead, combining it with a second bacteriostatic agent (e.g., NeoCide BND-10, BIT-10, GML-2, GML-3, GML-5) creates a multi-system synergistic effect, resulting in better inhibition and avoiding yellowing.

  5. PC-300 is not heat-resistant. For products requiring long-term high-temperature transportation, the combined preservative system described in item 4 is recommended.
